    Thesis Description:Much remains unknown about phospholipid transport in gram-negative bacteria, which plays a key role in maintaining the integrity of the bacterial outer membrane - a distinctive permeability barrier that confers resistance to external insults such as antibiotics.In my research, I employed a metabolic labelling strategy involving two synthetic probes - propargyl choline and 16-diazFA - to perform in vivo functionalisation of phospholipids biosynthesised in an engineered E. coli model system. Leveraging on UV cross-linking, click chemistry and affinity purification techniques, nine putative phospholipid-binding proteins were successfully captured. Subsequent characterisation and validation of candidate proteins will contribute to an improved mechanistic understanding of outer membrane biogenesis and lay scope for subcellular targets to be further investigated.Supervisors: A/P CHNG Shu Sin, Dr HOANG Truong GiangFinal Grade: A
